Egle Therapeutics, a clinical-stage biotechnology company advancing the next generation of regulatory T cell (Treg)-focused therapies for oncology and autoimmune diseases, today announced the appointment of experienced biotechnology industry executive, Christophe Quéva, Ph.D, as Chief Executive Officer (CEO). With over 20 years of experience in the biotechnology industry, Dr. Quéva brings a wealth of expertise as Egle transitions into the clinic its two lead drug candidates, EGL-001 and EGL-003.

Christophe Quéva, Ph.D.

Dr. Christophe Quéva brings over two decades of experience in immuno-oncology, spanning portfolio development from target identification to clinical trials and regulatory approvals. Before joining Egle, he founded and led Ovie Therapeutics, a company focused on an innovative treatment for brain cancer. Since 2022, he has also served as Chief Scientific Officer at DEM BioPharma, driving the development of cancer macrophage-targeting therapies. Prior to this, Dr. Quéva was Chief Scientific Officer at Oncorus, where he advanced next-generation immunotherapies using oncolytic viruses and RNA therapeutics. He also held the same role at iTeos Therapeutics, overseeing the development of a groundbreaking immuno-oncology portfolio.

Earlier in his career, Dr. Quéva held leadership roles at Gilead Sciences, Amgen, and AstraZeneca, contributing to oncology and inflammation drug discovery. He holds a Ph.D. in Life and Health Sciences from the University of Lille in France and completed postdoctoral research in cancer biology at the Fred Hutchinson Cancer Research Center. Dr. Quéva’s extensive career includes numerous patents and publications in the oncology field, reflecting his commitment to advancing patient care through innovative science and leadership.

Egle Therapeutics to share preclinical efficacy data for EGL-001, a CTLA-4/CD25 antagonist fusion protein at the Society for Immunotherapy of Cancer Meeting

Egle Therapeutics, a biotechnology company focused on advancing the next generation of regulatory T cell-focused therapies for oncology and auto-immunity, will present at the Society for Immunotherapy of Cancer Meeting taking place in Houston, Texas from November 6th to 10th, 2024.

The presentation, entitled “Preferential tumor retention of EGL-001, a CTLA-4/CD25 antagonist fusion protein, to selectively deplete tumor Tregs and minimize peripheral toxicities: towards a first-in-human clinical study”, will highlight the development and promising preclinical results of EGL-001, a humanized antibody designed to selectively target regulatory T cells (Tregs) within the tumor microenvironment. The data showcases EGL-001's potential as single-agent and to enhance the effectiveness of immune checkpoint blockade therapies by depleting tumor-associated Tregs, leading to increased CD8+ T cell activation and anti-tumor responses in preclinical models.

The SITC presentation will include key findings from studies in mouse models and cynomolgus monkeys, demonstrating the safety, tolerability, and efficacy of EGL-001, as well as its preferential tumor accumulation and rapid clearance from peripheral organs. These promising results leading Egle Therapeutics to conduct a first-in-human open-label, phase I/II trial evaluating the safety, tolerability, PK and preliminary activity of EGL-001 in patients with selected solid tumors, with the goal to address the critical need for more effective immunotherapies in cancer treatment.

Michel Detheux, Ph.D, Michel has held the role of Chief Executive Officer at iTeos Therapeutics since the company’s inception in 2012. More recently, he spearheaded the strategic collaboration with GlaxoSmithKline in 2021 to co-develop and co-commercialize iTeos’ differentiated anti-TIGIT antibody for a total value of $2B. He previously served as a director at Ludwig Cancer Research and worked in various scientific and business development roles at Ogeda (f/k/a Euroscreen). Dr. Detheux holds a degree of Bio-engineer and a Ph.D. in Biochemistry from Université Catholique de Louvain, and a business certificate from Solvay Business School.

Mojgan Hossein-Nia, Ph.D., Mojgan is the Head of R&D Global Portfolio Strategy and Strategic Finance, Research and Development at Takeda. She joined the company in September 2017 and currently directs portfolio strategy and management across all therapeutic areas facilitating investment decisions and overseeing R&D governance process. Additionally, she manages enabling capabilities that support portfolio management including the center of operational performance and integrated portfolio/finance digital solutions. In previous roles, she led the R&D investor relations group and built up the R&D Partnership Office, which was created to manage the overall health of R&D’s pipeline partnerships and their integration into R&D processes.

Before joining Takeda, Mojgan worked at Novartis Pharmaceuticals, building and leading multiple global functions across R&D. Mojgan’s career spans over 30 years, comprising dedicated roles in development outsourcing, global program leadership and project management, business development & licensing, and across multiple therapeutic areas including cardiovascular, metabolism, and cell & gene therapies.

Dr. Hossein-Nia holds a B.Sc, and Ph.D. in biochemistry from University of London, UK, and a diploma in pharmaceutical medicine from University of Surrey, UK.

Egle Therapeutics to share preclinical data for its first autoimmune disease program, EGL-003, with an oral presentation at the 2024 Promise of Interleukin-2 Therapy Meeting

Egle Therapeutics, a biotechnology company focused on advancing the next generation of regulatory T cell-focused therapies for oncology and auto- immunity, will deliver an oral presentation at The Promise of Interleukin-2 Therapy meeting taking place in Paris from September 4th to 7th, 2024.

The presentation, entitled “EGL-003, a novel IL-2 mutein to selectively expand and activate regulatory T cells and improve therapeutic efficacy in autoimmune disease”, will show evidence that EGL-003, a non-targeted Fc-fused IL-2 agonist mutein, selectively binds to regulatory T cells (Treg) and induces Treg IL-2 signaling.

The resulting increase of Treg frequencies was observed in vitro and in different mouse models. Accordingly, EGL-003 significantly improved clinical, macroscopic, and histological parameters in a DSS-induced colitis mouse model with expansion of tissue-resident Tregs.

EGL-003 is the first drug candidate being developed for treatment of autoimmune diseases in Egle’s pipeline and has advanced into IND-enabling studies.

Prior to Egle Therapeutics, Dr. Motlagh was Chief Medical Officer at Mablink, an ADC biotech company, where through the establishment of a differentiating clinical development plan he contributed to the value creation leading to an acquisition by Eli Lilly. Previously, he held global positions at leading oncology companies such as GSK, Bristol-Myers Squibb and AstraZeneca, where he was instrumental in the transition of several compounds towards the late clinical development phases of now approved drugs. John Celebi, MBA, brings over 25 years of experience building innovative entrepreneurial biotechnology companies. He currently serves as President and CEO of Sensei Biotherapeutics (NASDAQ: SNSE), a biotechnology company focused on the discovery and development of novel immunotherapies for cancer patients with unmet needs. Previously, Mr. Celebi served as the Chief Operating Officer of X4 Pharmaceuticals where he established and oversaw the company’s oncology business strategy. Egle Therapeutics to present novel immunocytokine preclinical data for EGL-001 with poster presentation at the 2024 AACR Annual Meeting

In the poster, entitled “EGL-001 is a novel immunocytokine designed to specifically target and disarm T regulatory cells in the tumor microenvironment”.

Egle scientists present evidence that EGL-001, a CTLA-4 targeted IL-2 dominant negative mutein, selectively binds to T regulatory (Treg) cells and inhibits Treg IL-2 signaling.

The resulting reduction of Treg frequencies and functionality, as observed in ex vivo human tumor cultures and in vivo mouse models, promotes effector T cell responses. Accordingly, EGL-001 induces anti-tumor responses in combination with anti-PD-1 treatment in mouse models.

EGL-001 is the first drug candidate to emerge from Egle’s discovery platform, and it’s being developed for treatment of advanced solid tumors and to restore responsiveness to PD-1 inhibition.

Dr. Gostissa has led preclinical programs at small and large biotech companies, focusing on how to best translate scientific insights into innovative cancer therapeutics. Most recently, she was Vice President of Preclinical Sciences at Jounce Therapeutics, where she contributed to establish the company’s scientific strategy and to oversee a broad preclinical portfolio of immuno-modulatory antibodies.

During her time at Jounce, Dr. Gostissa led efforts that enabled two IND applications, was involved in a successful outlicensing deal, and established and grew effective multi- disciplinary teams. Before starting her career in biotech, Dr. Gostissa spent many years in academic research, where she acquired a deep knowledge in cell biology, oncology and tumor immunology. Dr. Gostissa obtained her PhD in molecular genetics from the International School for Advanced Studies in Trieste, Italy, and completedpost- doctoral studies at Boston Children's Hospital/Harvard Medical School. She co-authored over 40 peer-reviewed publications and was the recipient of several awards, including a Leukemia and Lymphoma Society Special Fellow Award and a V Foundation Scholar Award.

Dr. Winter provides more than 10 years of experience in operationally leading and managing analytical & process development and external manufacturing activities. His expertise ranges from cell bank generation to drug substance and drug product manufacturing and supply.

Dr. Winter joined Egle Therapeutics in 2023, and assumes responsibility for the design, the establishment, and management of the manufacturing and supply strategy, as well as for the implementation of internal requirements and procedures to comply with applicable quality- related regulations and guidelines.

Previously, Dr. Winter worked as Head of CMC for a clinical-stage biotech company in Germany and as CMC Lead for Baxter/Shire PLC in Austria on numerous Investigational New Drug programs. In his past roles, he headed different teams to deliver and master all clinical trial-enabling CMC activities and actively contributed to numerous clinical trial applications and patents.

Dr. Winter is a trained biochemist and received his PhD in Natural Sciences from the Martin Luther University Halle-Wittenberg, Germany.

Egle Therapeutics raises 40 million € Series A
to develop First-In-Class T-regulatory cells therapies based on Treg-starvers

LSP and Bpifrance, through its InnoBio 2 Fund, co-led international investor syndicate

Egle Therapeutics SAS (Egle), an emerging biotechnology company focused on developing First-In- Class immunotherapies targeting immune suppressor regulatory T-cells (Tregs) for oncology and autoimmune diseases, today announces that it has completed a €40M ($46.4M) Series A financing. The Series A was co-led by LSP and Bpifrance through their InnoBio 2 fund. Fund+, Bioqube Ventures and Takeda Ventures, Inc. also participated in this round.

Egle Therapeutics was founded in early 2020 with a vision to become a game changer in the field of Tregs immunomodulation through the unique concept of Tregs starving and specifically targeting the most immunosuppressive ones. Spun out of Institut Curie, Egle’s scientific foundation leverages unprecedented, computational-based, IL-2 modified variants and newly tumor-associated Treg targets to build a furnished pipeline of First-In-Class immunocytokines against Tregs. The new capital will be used primarily to advance 2 leads assets into the clinic and further strengthen its internal drug pipeline.

The investor syndicate will join the Egle Therapeutics Board which will consist of Felice Verduyn-van Weegen (LSP), Vincent Brichard (LSP), Jean-Francois Morin (Bpifrance – InnoBio 2) and Sacha Mann (Takeda Ventures). Philippe Monteyne (Fund+), Jacques Mizrahi (Bioqube Ventures) and Elisa El Nouchi (Bpifrance InnoBio 2) will join as observers.

Egle to receive R&D milestone payment and equity funding from Takeda

Egle Therapeutics SAS (Egle), an emerging biotechnology company focused on developing first-in class immunotherapies targeting immune suppressor regulatory T-cells (Tregs) for oncology and autoimmune diseases, today announces that it has achieved the first milestone in its strategic research alliance with Takeda Pharmaceutical Company Limited (“Takeda”). In the course of the strategic research alliance that was announced in June 2020, Egle will validate novel tumor-infiltrating regulatory T-cell (Treg) targets against which Takeda will develop potential therapies.

Egle has leveraged its unique bioinformatic and translational capabilities to successfully identify targets that will now be pursued under the terms of the alliance. Upon achievement of the target identification, Egle will receive an R&D milestone payment and investment from Takeda.

Egle Therapeutics enters into a corporate strategic research alliance with Takeda to uncover novel tumor-specific regulatory T-cell targets and develop unique anti-suppressor-based immunotherapies

Egle Therapeutics SAS, an emerging biotechnology company focused on developing first-in-class immunotherapies targeting immune suppressor regulatory T-cells for oncology and autoimmune diseases, announced today a corporate strategic research alliance with Takeda Pharmaceutical Company Limited (“Takeda”) with the goal of validating novel tumor-infiltrating regulatory T-cell (Treg) targets against which Takeda will develop potential therapies.   

As a newly established bioscience company spun out of Institut Curie in the field of Treg immune modulation, Egle Therapeutics has assembled a proprietary translational based target discovery engine leveraging patient samples to map out - at the single cell level - unique transcriptomic signatures and targets specific for tumor-infiltrating Treg sub-populations. Capitalizing on these newly identified tumor-infiltrating Treg targets, the company is building a proprietary Treg modulating drug pipeline which also includes computationally designed resurfaced IL-2 proprietary variants, featuring unique mechanism of action to engage or dis-engage Tregs.

Under the terms of the agreement, Egle Therapeutics will lead target validation efforts on a subset targets Egle has identified and Takeda will be responsible for the development, manufacturing, and commercialization of resulting therapies. Egle Therapeutics will receive an upfront payment and research funding, and will be eligible to receive additional development and sales milestone payments based on the exclusivity of targets selected by Takeda.

In addition to the funding of the multi-target research collaboration, Takeda will invest €4.6 million in convertible debt through its venture arm, Takeda Ventures, Inc. (“TVI”), to support Egle’s corporate growth and internal programs on IL-2-based Treg modulation. Egle’s flagship program pioneers the dis-engagement of tumor-infiltrating Treg through unique proprietary series of IL-2 variants that act as IL-2 antagonists.

“While the whole competition converges solely on a similar approach to enhance effector T cells by disrupting binding of IL-2 to Tregs, our IL-2 variants conserve intact IL-2 binding to CD25, conferring a unique mechanism of action for starving Treg from endogenous IL-2, while leaving active the CD8+ effector T cell arm of the immune response,” Luc Boblet added. “Takeda’s equity commitment will help Egle’s launch strikingly hit the road building on our excellent scientific foundation.”
